Eli Lilly to Present New Oncology Data at ESMO Congress 2026

Eli Lilly’s oncology pipeline will feature at ESMO Congress 2026, with data presentations on investigational sofetabart mipitecan and olomorasib, alongside updated findings for Retevmo (selpercatinib) and Verzenio (abemaciclib).

Eli Lilly to Showcase Oncology Pipeline Advances at ESMO Congress 2026

Eli Lilly will present new data from across its oncology portfolio at the European Society for Medical Oncology (ESMO) Congress 2026, scheduled to take place from October 23 to 27 in Madrid, Spain.

Key highlights include Phase 1 data on sofetabart mipitecan (Sofe-M), the company’s investigational folate receptor alpha (FRα)-targeting antibody-drug conjugate (ADC), in combination with bevacizumab for platinum-sensitive ovarian cancer (PSOC). The company will also present updated findings on olomorasib, Retevmo (selpercatinib) and Verzenio (abemaciclib) in lung, thyroid and breast cancers, respectively.

“Lilly’s presentations at ESMO reflect both the breadth of our oncology portfolio and the focus behind it: following compelling science wherever it has the potential to change care,” said Jacob Van Naarden, executive vice president and president of Lilly Oncology.

He added that the data on sofetabart mipitecan in platinum-sensitive ovarian cancer build on earlier findings in platinum-resistant disease and mark a key step in advancing the ongoing Phase 3 FRAmework-01 study across both settings.

Van Naarden also described the updated first-line data for olomorasib in KRAS G12C-mutant lung cancer as promising, citing clinical activity and a safety profile consistent with earlier findings.

“Alongside new data in thyroid and breast cancers, these presentations represent meaningful momentum toward improving care for people with cancer,” he said.

Sofetabart Mipitecan

Sofetabart mipitecan is an investigational ADC comprising an Fc-silent, humanised monoclonal antibody targeting FRα, linked to exatecan, a topoisomerase I inhibitor, through a proprietary cleavable polysarcosine linker (PSARlink).

The drug is designed to target FRα across different expression levels, with the aim of improving its therapeutic index. FRα is a cell-surface glycoprotein encoded by the gene FOLR1 that binds to the essential nutrient folic acid and reduced folates, bringing them into cells to facilitate cell division and growth. FRα is overexpressed in many solid tumours such as ovarian and endometrial cancers.

Sofetabart mipitecan has received Breakthrough Therapy Designation from the US Food and Drug Administration (FDA) for platinum-resistant ovarian cancer (PROC) and is being evaluated in the Phase 3 FRAmework-01 trial.

At ESMO Congress 2026, Lilly will deliver a rapid oral presentation featuring the first Phase 1 data on sofetabart mipitecan in combination with bevacizumab in patients with PSOC.

Olomorasib

Olomorasib is an investigational, oral, potent and highly selective second-generation inhibitor of the KRAS G12C protein. KRAS is one of the most frequently mutated oncogenes across cancer types. KRAS G12C mutations occur in approximately 13% of patients with non-small cell lung cancer (NSCLC) and 1–3% of patients with other solid tumours.

The drug is being studied in combination with pembrolizumab, with or without chemotherapy, as a potential first-line treatment for advanced NSCLC with KRAS G12C mutations. It is also being evaluated alongside immunotherapy in resected and unresectable NSCLC, as well as alone and in combination regimens for other advanced solid tumours.

At the congress, Lilly will present an updated integrated analysis from the Phase 1/2 LOXO-RAS-20001 trial and the safety lead-in portion of the Phase 3 SUNRAY-01 trial. The analysis assesses olomorasib in combination with chemoimmunotherapy comprising pembrolizumab, pemetrexed and platinum-based chemotherapy as a potential first-line treatment for patients with advanced or metastatic KRAS G12C-mutant NSCLC.

Retevmo

Retevmo (selpercatinib), formerly known as LOXO-292, is a selective and potent RET kinase inhibitor with central nervous system activity. The drug is approved by the US FDA for the treatment of advanced or metastatic medullary thyroid cancer (MTC) with a RET mutation in adults and children aged two years and older who require systemic therapy.

Lilly will present updated results from the Phase 3 LIBRETTO-531 study, which evaluated selpercatinib against cabozantinib or vandetanib in patients with advanced, RET-mutant MTC who had not previously received a kinase inhibitor. The results have supported global approvals of Retevmo for this treatment setting.

Verzenio

Verzenio (abemaciclib) is an oral cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor approved for certain patients with hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2−) breast cancer in the adjuvant and advanced or metastatic settings.

First approved in 2017, the drug is authorised for use in more than 90 countries around the world.

At ESMO Congress 2026, Lilly will present overall survival data from the Phase 3 post-MONARCH trial, which evaluated abemaciclib plus fulvestrant in patients with HR-positive, HER2-negative advanced breast cancer whose disease progressed following prior CDK4/6 inhibitor and endocrine therapy.

According to Lilly, the findings complement results from the EMBER-3 trial evaluating abemaciclib plus imlunestrant and add to the evidence supporting a change in endocrine therapy alongside CDK4/6 inhibition following disease progression in metastatic breast cancer.
